While the\nthree-dimensional stru ctures of functional proteins are determined\nby their amino acid sequence s\, as originally demonstrated by\nAnfinsen\, it has become clear that wit hin the crowded environment\nof cells many proteins depend on so-called mo lecular chaperones to\nreach their folded states efficiently and at a biol ogically\nrelevant time scale. Assistance of protein folding is provided b y\ndifferent types of chaperone which act to prevent misfolding and\naggre gation\, often in an ATP-dependent mechanism. In the cytosol\,\nnascent ch ain-binding chaperones\, including Trigger factor and\nHsp70\, stabilize e longating polypeptide chains on ribosomes in a\nnon-aggregated state. Fold ing is then achieved either on controlled\nchain release from these factor s or following polypeptide transfer\nto downstream chaperones\, such as th e cylindrical chaperonins GroEL\nand TRiC. The latter provide nano-compart ments for single protein\nmolecules to fold in isolation\, unimpaired by a ggregation.\nOnce folded\, many proteins continue to require chaperones to \nretain their functional states\, particularly under conditions of\ncell stress. Failure of the chaperone machinery to maintain\nproteostasis\, i.e . the conformational integrity of the cellular\nproteome\, may facilitate the manifestation of diseases in which\nproteins misfold and are deposited as aggregates\, such as\nParkinson’s and Huntington’s disease. A decline in proteostasis\ncapacity occurs during aging\, presumably explaining why age is the\nmajor risk factor of neurodegenerative diseases.
