Her group focuses on the identification an d\nfunctional characterization of protein interaction modules\nsurrounding serine/threonine phosphatases. Active areas of research\ninclude understa nding how the PP4 phosphatase modulates gene\nexpression via regulation of transcription elongation\, mRNA\ncapping\, and splicing. The Gingras lab is also investigating the\nfunction of a novel protein complex that they h ave identified\nsurrounding the protein CCM3\, whose gene is mutated in fa milial\ncases of angioma.  Anne-Claude Gingras is also involved in the\nde velopment of robust approaches for the analysis of protein\ninteraction ne tworks in yeast and human and for the study of\nenzyme-substrate relations hips within the interaction\nnetworks.\n \nProtein phosphorylation mediate s cellular responses to growth\nfactors\, environmental signals\, and inte rnal processes by the\nregulation of protein interactions\, enzyme activit y or protein\nlocalization. However\, the protein interactions of kinases\ ,\nphosphatases\, their regulatory subunits and substrates remain\nsparsel y mapped. Two interaction mapping projects will be\npresented: 1) To chart the budding yeast kinase and phosphatase\ninteraction (KPI) network\, we systematically characterized protein\nkinase and phosphatase complexes by sensitive affinity purification\ncoupled to mass spectrometric identificat ion (AP-MS).  We\nidentified a KPI network of 1\,844 interactions that con tained many\ndense local regions.  Notably\, the cell cycle phosphatase\nC dc14 associated with multiple kinases that revealed roles for\nCdc14 in mi togen-activated protein kinase signaling\, the DNA damage\nresponse and me tabolism\, while interactions of the target of\nrapamycin complex 1 (TORC1 ) uncovered new effector kinases in\nnitrogen and carbon metabolism.
