Duke Professor\nMolecular Genetics & Microbiology\nDuke University Medical Center\n“Coordinated Dynamics of RN A Regulation by RNA-Binding\nProteins”\nWhile RNA-binding proteins (RBPs) coordinate many key decisions\nduring cell growth and differentiation\, th e dynamics of RNA\nstability and translation that depend upon RBPs have no t been\nstudied on a global basis. Recent findings demonstrate that\nmulti ple mRNAs are co-regulated by sequence-specific RBPs that\norchestrate the ir splicing\, export\, stability\, localization and\ntranslation. These ob servations have given rise to a model in which\nmRNAs that encode function ally related proteins are coordinated\ndynamically during cell growth and differentiation as\npost-transcriptional RNA operons or regulons. I will r eview\nrecently discovered examples of post-transcriptional RNA operons in \nmammalian cells\, trypanosomes and budding yeast\, and their\npotential importance in processes such as immune response\,\noxidative stress and di sease. I will demonstrate how quantitative\nRNA dynamics involving the HuR RBP can be assessed\nprobabilistically using the RIP-chip method to deriv e a\nquantitative phenotype from the drug-gene-phenotype Connectivity\nMap that allows us to identify small molecule drugs affecting\nposttranscript ional regulatory networks during T-cell activation.\nIn addition\, I will discuss the use of the novel PAR-CLIP approach\nof Tuschl and coworkers to identify precise HuR RBP binding sites\nand how those data can complement RIP-chip data to visualize\ncoordination of global RNA dynamics affecting distinct functional\noutcomes.\nMorris\, A. et al.
